Try to visit early in the day or save the duomo for later, after all the tour groups have left so you can enjoy the interior with some peace. You can't miss the Duomo in Florence. The line to get inside the cathedral was a bit long, but it moved very fast. The art and architecture inside was just amazing to behold. Don't forget to visit the Duomo museum across the street. more

Obviously, if you visit Florence you'll see the Duomo. If possible, go inside. Entrance is free. You need sensible cloth like in any italian church. Unfortunately, tourists can only wander on the back half of the cathedral, without chairs; and it's unpractical for looking at some paintings.
